The E10S eyepiece with 10x magnification has a graduated scale on the plane of the field diaphragm. It acts as a regular eyepiece with 10x magnification, and is also capable of measuring the size of objects or related elements. The scale is marked in millimeters and centimeters. To measure, you must first calibrate the eyepiece for each objective you want to use. The E10S eyepiece fits into a 23.2mm tube.

Calibration

To perform calibration, you will need a calibration slide with a division of the scale of 0.01 mm. Place the slide on the stage and point the microscope at the scale. Then, rotate the eyepiece in the tube until the scale divisions of the slide and eyepiece are parallel to each other. Finally, make a calculation.

Calculation

It is necessary to calculate the relationship of the divisions on the scale of the eyepiece and that of the calibration slide (table micrometer ): how many divisions of the eyepiece scale fit into the scale of the slide (for low magnification objectives) or how many divisions on the slide fit into the eyepiece scale (for medium or high magnification objectives). Then use the formula:
E=TL/A, where:
E is the eyepiece scale division;
T is the stage micrometer scale division marked on the stage micrometer ( 0.01 mm);
L is the number of values on the stage micrometer;
A is the number of values on the eyepiece scale.

Record the number obtained from the calculations for each objective.
The size of the samples is calculated as follows: it is necessary to determine how many divisions the sample occupies on the eyepiece scale. Multiply this number by the result obtained from each objective's formula. This number will reflect the linear value of the sample.
